L-Asparaginase: A Promising Enzyme for Treatment of Acute Lymphoblastic Leukiemia
نویسندگان
چکیده
Asparaginases are known to be the cornerstone for treatment of acute lymphoblastic leukemia (ALL) and are used for treatment in all pediatric regimens as well as in the majority of adult treatment protocols. Clinical hypersensitivity reactions against commercially available asparaginase have resulted in failure of asparaginase in treatment of ALL in more than 60% of cases. Thus, it is required to search for serologically different asparaginase from new organisms for the patients exhibiting sensitivity to one formulation of asparaginase, so that they can be switched to another to ensure that they receive the most efficacious treatment regimen possible. The present study report E. coli VRY-15, E. coli VRY-8 and E. coli VRY-14 as potent producer of L-asparaginase. The L-asparaginase obtained from E. coli VRY-15 showed highest specific activity i.e., 19.56 μmol/mg. Attempt was made to purify the enzyme. Molecular weight of purified L-asparaginase obtained from E. coli VRY-15 was found to be 56 K Da as determined using Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDSPAGE).

Isolation and identification of L-asparaginase producing actinomycetes from Persian Gulf
Introduction: L-asparaginase is an anti-neoblastic agent used in the chemotherapy of lymphoblastic leukemia. This enzyme is widely distributed among microorganisms, plants and animals, but microorganisms have proved to be a better alternative for L-asparaginase, thus facilitating its large-scale production. Actinomycetes are filamentous, Gram-positive bacteria widely distributed in the marine e...

Introduction: The bacterial Asparaginase is used in the treatment of asparagine-dependent tumors, particularly lymphatic sarcoma and acute lymphoblastic leukemia. However, the instability of the enzyme increases the number of injections that are accompanied by high immune responses. The aim of this study was to investigate the conjugation of L-asparaginase with nanochitosan glutaraldehyde (NCG)...

Backgroung and Aims: L-Asparaginase II is a cornerstone of treatment protocols for acute lymphoblastic leukemia. Only asparaginase II obtained from E. coli K12 and Erwinia chrysanthemi have been used in human as therapeutic drug. The therapeutic effects of asparaginase II from E. coli K12 and Erwinia chrysanthemi is accompanied by side effects. It is desirable to search for other asparaginase I...

Background: L-Asparaginase (L-Asp) is used as an efficient anti-cancer drug, especially for acute lymphoblastic leukemia (ALL). Currently, two bacterial asparaginase isoenzymes are used for cancer treatment. Therefore, this research focused on isolating native bacteria with the ability to produce L-Asp. Materials and methods: L-Asp producing bacteria were isolated from soil samples on 9K medi...
